| Recombinant Human FGF Basic
Source: The human gene sequence was expressed in E.coli and the protein purified by sequential chromotography.
Purity & Assay Method: >97%, as determined by SDS-PAGE, visualized by silver stain
Formulation: Lyophilized from a 0.2 µm filtered solution in 20 mM Tris HCL, pH 7.0, containing 50 µg of bovine serum albumin per 1 µg of cytokine.
Mass Per Vial: There are 3 vial sizes, 10 µg, 100 µg, and 1000 µg of recombinant human FGF Basic
Activity & Assay Method: The biological activity of recombinant human FGF basic was monitored in a mitogenic assay by measuring the FGF basic dependent3 [RH-1]H-thymidine incomporation in a quiescent NR6R firbroblasts (Rizzino, A. et al., 1988, Cancer Research 48:4266-4271; Thomas, K., 1987, Methods in Enzymology, 147:120-135). The ED50 for this effect is typically 0.1-0.25 ng/mL.
Reconstitution: Reconstitute with sterile PBS containing at least 0.1% HSA or BSA and 1mM DTT. The rhFGF basic concentration in the stock solution should be no less than 10 µg.
Storage Conditions: Lyophilized samples are stable for greater than six months at -20ºC to -70ºC. Upon reconstitution, this cytokine can be stored under sterile conditions at 2º to 4ºC for one month or at -20ºC to -70ºC for three months without detectable loss of activity. AVOID REPEATED FREEZE-THAW CYCLES.
- Sterilization: FGF Basic was filtered through a 0.2 micron membrane and packaged under aseptic conditions.
Endotoxin Level & Assay Method: The endotoxin level was determined by the LAL method and found to be less than 0.ng per 1µg of the cytokine.
CAUTION:FOR RESEARCH USE ONLY. NOT FOR DRUG OR DIAGNOSTIC USE. NOT FOR USE IN HUMANS.
